At a party you should stick to the fun stuff to complement the fun environment of a party....do routines, games, palm reading, tests (e.g. best friends test, the cube), basically anything thats fun.
If you feel like you're having success, don't push your luck too far...grab that number. Possibly arrange to meet up later that night, but don't stay with the same chick the whole time if you're at a party. She wants to talk to other people, you want to pickup more chicks. its a win-win.

Don't make yourself too available you'll get into the "Lets just be friends" zone if you're too comfortable. Instead, talk to other groups, let her see you having a great time, talk to other women this way she'll feel like she's competing, and if possible go somewhere else and keep your game on!
