There is that line in Mystery Method that says "If I weren't gay, I'd be all over you" or something like that. You have to do what she doesn't expect, and you have to show that you don't give a shit if she is into you or not. You don't care what she thinks of you.
Her: Are you gay or something?
YOU: Yeah, actually. I'm going to pride next week. The parties there are great! But you wouldn't know about that, would you.
Her: Oh... Uh, why wouldn't I know about that? / What do you mean?
YOU: Well, your kinda killing the mood right now. I wouldn't party with you based on that attitude, come on, lets get serious.

As the conversation goes on, play with her a bit, use all the principles of game, build attraction and rapport and escalate. Eventually the gay thing will come up again, then you can just tell her you were kidding, or you went straight for her, just for this night, or something. Be creative.
If she has a negative reaction...
Her: Are you gay or something?
YOU: Yeah, I am. blah blah. Blah. / What if I am?
Her: Oh, thought so. [dirty look] / giggles and tries to walk off / turns around / etc.
YOU: Why, does that matter to you? / Oh, so you were interested, then? / Don't be so shallow.
