If the target ask's you while in the middle of the game 'Aren't you doin' this thing called 'The game'?
What should you reply?

Plausible deniability. You can just act like you have no idea what she's talking about "What 'game'? I'm good at tic tac toe. Want to play?". If she follows up with an explanation, say "Oh that sounds really cool. I'm going to have to try it! Does it work?"

Alternatively, you can say yes without saying yes directly. "Well, it seems to be working."

I really like the "it seems to be working" line

Normally i just go "Yes, isn't a guy with game better than someone who does not?" in a nonchalant way

You can use something like this:

Her: ''Are you doing The Game.''
You: ''Hell yeah I'm doing it and I should warn you, I'm pretty good at it, I always win.''
Her: ''Yeah right/I don't care/my hair's on fire''
You: ''No really, I've been playing it since childhood and once I've lost the cards and the figures and I had to buy it again.''
Her: ''Huh/What ?''
You: ''I thought you were talking about the Monopoly ''

I haven't field tested it though, cause I was never called on using game

When called out I tend to throw out a playful, teasing disqualfier, or absurdify what she said to the point of it being ridiculous.

Her: "Are you doing the game."
Me: "Wowww. You're really cocky. What makes you think I'd try to run game on you. I'm trying to get at your friend."
Or.
"Game? I'm so awful with woman it's ridiculous."
Or
"You're a smart one. You might be a keeper. Too bad you ain't my type. If you were half black and half asian that weighed 350 lbs we might be able to make something work."

Saying something along those lines seem to work with for me.

This is really a shit test, and a lot of people think they're bad, but in reality they are good. A girl is testing you because she's attracted to you and she wants to make sure you're all that you seem.
