I'm looking for advice on letting girls down you're in the midst of picking up: after either 1) they've said/done something which put up red flags that you just don't want to deal with, or 2) other opportunities come into the establishment you want to pursue...

This is mostly in the context of bars/clubs/parties where you will likely be in the same room as the girl you were originally trying to pick up.

How do you play this?

Is there any possible way of doing this without being a total dick?

If you're thinking, "well, who cares if you seem like a dick?" I could give specific scenarios of why a clean extraction from the situation would be desirable. Hopefully, though, you get where I'm coming from.

Simple, two methods I would take into consideration is Mystery's Method of Rejection or Adam Lyon's (AFCADAM) rejection theory. Both work in similar ways but Adam Lyons is more subjected to the UK's norms. Mystery's Method is to just brush them of and say "Haha *don't emphasise the laugh* ok nice meeting you" (if you build social proof at the end of the night then they'll look like idiots, not vice versa). Adam Lyons has a little more depth into it, if he gets rejected he would say "Ok, sorry for bugging you *sigh* nice meeting you" and the same method applies, if you build up social proof later they'll look like total idiots, not you.
