So I messaged this one chick on Pof and said "omg you look like one of those girls that needs a days notice in to hangout because they take so long to get ready. Ill give you a shot anwayway. Check out my profile and hit me back". She replied with "hey there, what's up!" Where can I take it from there?

This is why I like online sarging at times. It gives you a challenge on how to carry conversations on and on.
She responded, so that's a good thing.
Just explain something insignificant in an exciting way "I'm fighting not to eat this snickers bar but it's so tempting.." then switch the conversation back to herself, so she can open up more.

If nothing else, I just say "So tell me about yourself" and 9/10 they explain their interests and you can go from there. Think of it as sharpening your conversational skill.

That's not the most promising of responses but it's a response. Generally, I try to qualify them somehow and get them on the phone or out on a date pretty quickly. It's not all that high probability with a "What's up?" but working on building attraction over time with chicks on a dating website is usually a waste of time.
